This fact has been known since the development of the first engines in the late 1800´s. As fossil fuels are a limited source of power, more sustainable ways of energy supply need to be investigated, tested and implemented. Alternative fuels such as Biodiesel have been investigated and used for more than 30 years, especially in Austria.This master thesis focuses on the use of non-fossil diesel fuels for powering construction machines such as Excavators, Wheel Loaders and Off-Highway Trucks. Those machines are mostly powered by diesel engines and as they consume huge amounts of fuel while moving earth, they are an interesting area of improvement. The author drew the conclusion that all construction machines, powered by diesel engines, can be run on pure Biodiesel when particular application parameters are met and the consequences are taken into account. Furthermore the author encourages the representatives of the earthmoving business to perform field tests for proving the suitability of Biodiesel for the use in diesel engines. This could furthermore lead to a reduced dependence on fossil diesel in this industrial sector.

The increased industrialization and motorization has led to a steep rise for the demand of petroleum products. Petroleum based fuels are obtained from limited reserves. It is estimated that the known crude oil reserves could be depleted in less than 50 years at the present rate of consumption. Hence, it is necessary to look for alternative fuels. An entire branch of biotechnology, referred to as "white biotechnology", centers on the bio production of fuels and chemicals from renewable sources. About 90% of the current biofuel market is represented by biodiesel and bioethanol. Biodiesel is an efficient, nontoxic, biodegradable and clean burning fuel alternative to petroleum fuels which runs in unmodified diesel engines. This paper reviews approaches to microbial synthesis of biodiesel, focusing on the role of synthetic biology as an enabling technology in the design of optimal cell factories.

GREET enables a fast approach to an optimum Well-to-wheel analysis, while bringing new insights and improving the understanding of the nature of various biodiesel production processes. A simulation of Well-to-wheel analysis of Bio-diesel from soyabean is carried on GREET is discussed in this book. These simulation mentions about various steps involved in the analysis. It is followed by detailed tabulation of the analysis. Furthermore Life Cycle Analysis of Bio-diesel production from jatropha and micro algae is developed and discussed. In this book, Net Energy Ratio (NER)of these bio-diesel sources is calculated and a comparison with production of conventional diesel is made. Thus, the main aim of this book is to inform scholars about GREET and its effective use in industries as well as by research community and to assess the sustainability of Bio-diesel production from jatropha and micro algae and their comparison with conventional diesel.

(Biotechnology), language: English, abstract: Abstract: Jatropha curcas will be a vast source of biofuel and a key to reducing our dependence on fossil fuels. Various government agencies around the world have proposed production of biodiesel as a renewable alternative to fossil fuel. Jatropha curcas is frequently mentioned as the best option for producing biodiesel. Despite of having potential as an alternative fossil fuel, J. curcas is not being fully exploited. Hence, there is a need to identify high yielding clones of J. curcas for its further improvement. Molecular marker analysis in genome studies enhance the speed and efficiency of crop improvement, for that we need the protocols of DNA isolation; which is used to obtain high quality and quantity of DNA. Here the objective is to carry out comparative analysis of nine different DNA isolation protocols, from those only six methods were able to isolate DNA from such secondary metabolite producing plant, J. curcas. The most effective method of DNA isolation is modified CTAB method. The extracted DNA from the leaves of Jatropha curcas had 83% purity. The extracted DNA was found suitable for restriction digestion and for different DNA fingerprinting techniques like, RAPD; which helps for the further improvement of plant. They can be divided in first generation and second generation biofuels. First generation biofuels have an inefficient CO2 emissions to output ratio. In fact they currently need mineral-oil for their production. Second generation biofuels have nearly the same energy efficiency with much lower environmental damages. Producing a second generation biofuel, for example from algae, most of the needed process energy can be produced by the process itself. CO2 outputs will be recycled by using it as an input product. To get a high biofuel yield from algae, light availability and intensity must be optimized to the current algae-species. Therefore biofuel reactors need to be optimized for a homogeneous light saturation. To estimate the innovation potential of biofuels from algae nine different assessment techniques where used. The result is, that there is no main method for biofuel production from algae on an industrial scale. It depends on consumer requirements, location and the technology.

Biodesulfurization (BDS) of fossil fuels has emerged as a promising alternative to the conventional hydrodesulfurization process for containment of sulfur oxide emissions during fuel combustion. The 4S pathway is a sulfur specific pathway that removes the sulfur atom from the recalcitrant organosulfur compounds present in fossil fuel fractions like diesel by oxidation. The end product is a sulfurless molecule, 2-hydroxybiphenyl. Though the process is very specific, however, speculations as to the bacterium's organic solvent tolerance for effective BDS process are justified. This book focuses on the construction of genomic library of a biodesulfurizing bacterium in an organic solvent bacterium in an effort to enhance biodesulfurization of a model compound dibenzothiophene, by the recombinant bacterium.

It is estimated that ca. 16% of the world's agro-climatic areas are affected by waterlogging. It causes significant reduction in the yield of several crops. Jatropha curcas L. is a deciduous shrub, drought-resistant but prone to waterlogging, oil bearing plant species, widely distributed in tropical and sub-tropical areas viz., in Central and South America, Africa, India and Southeast Asia. The plant produces useful products particularly seeds from which oil can be extracted (~35%), may be used in soap and cosmetics industries, and also as an important substitute for diesel and kerosene. Accordingly, it is expected that by 2015, Jatropha may be planted on 12.8 mha land globally for blending of fossil diesel with biodiesel across the world under climate change to mitigate green house gases (GHGs). An improved carbon assimilation through photosynthesis to achieve enhanced net plant produce and oil content is the need of future. Therefore, an agro-profitable cultivation of Jatrpoha be recommended with careful crop management through precision irrigation in years to come to enhance plant performance and seed yield to sustain bio-fuel production, bio-refining and bio-economy

The world's ever-growing demand for power has created an urgent need for new efficient and sustainable sources of energy and electricity. Fuel cells are an important alternative energy source, with promise in military, commercial and industrial applications, for example power vehicles and portable devices. A fuel cell is an electrochemical device that directly converts the chemical energy of a fuel into electrical energy. Fuel cells represent the most efficient energy conversion technologies to-date and are an integral part in the new and renewable energy chain (e.g., solar, wind and hydropower). Fuel cells can be classified as either high-temperature or lowtemperature, depending on their operating temperature, and have different materials requirements. This book is dedicated to the study of high temperature fuel cells. In hightemperature fuel cells, the electrolyte materials are ceramic or molten carbonate, while the electrode materials are ceramic or metal (but not precious metal). High operation temperature fuel cells allow internal reforming, promote rapid kinetics with non-precious materials and offer high flexibilities in fuel choice, and are potential and viable candidate to moderate the fast increase in power requirements and to minimize the impact of the increased power consumption on the environment. Fossil fuel accounts the highest percent of primary energy consumption and used extensively for transport and other purposes since 1859. Continued use of fossil fuel are recognized as unsustainable because of depleting world reserves and the green house gas emissions associated with their use. Therefore, study on microalgal biofuel was performed to evaluate the potential of algal biomass as biofuel resources. Also, it is focused on current methods for culturing, harvesting and extracting technology of microalgal biomass and economic, energetic and environmental impact of biodiesel production. Calculation of total energy yield per hectare per year with oil content of 30% dry weight microalgae was calculated with the values obtained from literature. Also, production of biodiesel per hectare per year was calculated and compared with other studies. Results obtained in our study showed similar pattern with other study. From our study, we found that total oil yield from algal biomass is much higher than terrestrial plants and therefore, microalgae have the potential to completely replace transportation petrol fuel.
