The fungus Ashbya gossypii is an important industrial producer of riboflavin, i.e. vitamin B2. Here, we developed and then used a highly sophisticated set-up of parallel 13C tracer studies with labeling analysis by GC/MS, LC/MS, 1D, and 2D NMR to resolve carbon fluxes and obtain a detailed picture of the underlying metabolism in the overproducing strain A. gossypii B2 during growth and riboflavin production from a complex industrial medium using vegetable oil as carbon source. Glycine was exclusively used as carbon-two - but not carbon-one (C1) - donor of the vitamin's pyrimidine ring due to the proven absence of a functional glycine cleavage system. Yeast extract was the main carbon source during growth, while still contributing 8 % overall carbon to riboflavin. Overall carbon flux from rapeseed oil into riboflavin equaled 80 %. Transmembrane formate flux simulations revealed that the C1-supply displayed a severe bottleneck during the initial riboflavin production, which was overcome in later phases of the cultivation by intrinsic formate accumulation. The transiently limiting C1-pool was successfully replenished by time-resolved feeding of formate or serine. This increased the intracellular precursor availability and resulted in a riboflavin titer increase of 45 %. This study is the first that successfully sheds light on carbon fluxes during the growth and riboflavin production phase by use of 13C tracers and a complementary platform of analytical techniques.

This book presents the findings of a research study on boys with behavioural difficulties, aged 5 to 7 years old. Boys with and without behaviour problems were told several stories with ethical dilemmas using lego-type dolls. They were asked to continue and finish each story in their preferred way. The children's behaviour was assessed in terms of aggression, intentionality and avoidance using a scoring system. The scores were analysed using Structural Equation Modelling to interpret the complex inter-relationships in their behaviour and to test the effect of demographic characteristics. The results indicate that there was an increased risk of anti social behaviour when avoidance and aggression were demonstrated in the same stories. Additionally, boys with more aggressive behaviour during the stories were more likely to demonstrate lower intentionality. This publication is written for those interested in the psychology of boys with behavioural difficulties and those with an interest in the application of Structural Equation Modelling in health research.

During the production life of oil wells, water is produced from the producer well due to the encroachment of water from aquifer or injected water in the reservoir, and sometimes the volume of water produced is much higher than the volume of oil produced. However, water can play an important role in maintaining reservoir pressure; it needs to be shut off when its' percentage reaches uneconomical limits. Excessive water production in oil fields results in a restriction of oil production and creates several production problems, including high lifting cost, producing corrosion and scale in production facilities and even sand production. In addition, high costs are incurred to separate, treat and dispose of the produced water. Produced water also contains organic and inorganic minerals which have a substantial negative impact on the environment.

The early quantitative characterization of possible production hosts is a key element in the development of knowledge based biotechnological processes. Despite of scale independent parameters like substrate uptake rates, production rates, maximum growth rates and yields, the performance on mixed substrates, as they occur for example during induction of recombinant protein production, is of great interest. In the present work, pulse and shift experiments with glycerol, glucose, malate, pyruvate and acetate were performed in chemostat cultures of the methylotrophic yeast Pichia pastoris. This allowed the quantitative examination of mixed substrate dynamics and the adaptation to higher growth rates. Different on- and off-line methods were used for a quantitative monitoring of the fermentation process and as input variables to mass balances. Elemental balances in combination with a statistical test for validation of the results were used for data reconciliation. Based on the results, the author discusses the pulsed substances towards their repressive, additive or competitive behaviour as mixed substrates and their effect on the yeasts metabolism.

Fruits and vegetables are important component in agriculture sector. Production and marketing studies were used to explore the cost parameters of the production and marketing and understand the inefficiency during production process and marketing channels. To observe the inefficiency during production, stochastic frontier production was used by using the primary data from two Tehsils of the Sargodha region known as Pakistan's California. It was concluded that all the input variables was directly related to technical efficiency except irrigation. Negative sign of the irrigation showed inverse relationship between irrigation and technical efficiency due to intercrop irrigation which resulted in over irrigation. Technical inefficiency model showed a positive relationship between age of the farmers, years of education and source of credit (dummy variable) while age-squared is inversely related to technical inefficiency. The marketing studies showed that by decreasing the number of the intermediaries in both form i.e. open and packed, not only the purchased price of the consumer decreased but also marketing efficiency of the market increased.

Master's Thesis from the year 2016 in the subject Business economics - Supply, Production, Logistics, , course: Agricultural Economics, language: English, abstract: This study aimed to analyze the technical efficiency of sesame production in Humera area and to identify major factors that cause efficiency differentials of smallholder farmers. The objective of the study is to measure the technical efficiency of small holder farmers in sesame production. The study was conducted using a cross sectional data collected in 2015/2016-production year from a total sample of 110 households. Cobb-Douglas function was employed to estimate technical efficiency of smallholder farmers in sesame production. The finding of the study indicated that there is inefficiency in the production of sesame in the study area. The estimation of the frontier model with inefficiency variables shows that the mean technical efficiency of farmers is 0.69 (69%). This implies that production of sesame can be increased by 31 percent given the existing technological level. This indicates that the farmers did not using production inputs efficiently in such a way that they give their maximum potential. The estimated stochastic production frontier model together with the inefficiency parameters suggests that any attempt to strengthen technical efficiency of smallholder farmers in the study area must give due attention to the improvement of the principal causes for efficiency differentials such as education, age, ...

Market-makers seem to provide less liquidity recently. The reasons of such a behaviour are shown within this work. It exhibits the regulations which have changed, the behaviour of market-makers and how financial markets are able to become illiquid. After this more theoretical framework, which refers to financial stability, several measures of liquidity are introduced and empirically tested on a dataset of about 60,000 corporate and sovereign bonds in 34 countries over a period of eleven years. The result, is that bond markets became less liquid within the last three years than during the financial and the following European debt crisis.

The expression of positive emotion, even though it is expressed more subtle, is no less important. This study was therefore conducted with the objective to identify, elicit and measure behavioural and physiological indicators of positive emotion in pigs. With the ability to facilitate a positive emotional state, the welfare of pigs kept in intensive livestock production systems can be greatly improved.
