The mucoadhesive drug delivery system has received great deal of attention over the past few decades. The major advantages of this system are to prolong the drug residence at a specific site of delivery and solve bioavailability problems. The major focus of this book is to explain the current research tools for designing of mucoadhesive microparticles of rabeprazole sodium and tolterodine tartrate by non-aqueous solvent evaporation technique using ethylcellulose and selected mucoadhesive polymers. The prepared microparticles of the above drugs were characterized for particle size, drug content, swelling index, mucoadhesive strength etc. In-vitro drug release study for both drugs suggested that formulation containing tolterodine tartrate with ethylcellulose based chitosan and rabeprazole sodium with ethylcellulose based HPMC K100M showed consistent drug release up to 24h. Accelerated stability study of optimized formulations such as F4 and F6c up to 6 and 3 months respectively.

Sublingual drug delivery has now been recognized as a very promising route for delivery of therapeutic compounds including biopharmaceuticals. The sublingual mucosa offers a large, very thin, highly vascularised layer for efficient absorption of drug. Buspirone hydrochloride gets completely absorbed from the gastrointestinal tract after oral doses but undergoes extensive firstpass metabolism with a bioavailability of about 5%. Till date, pursuit for cost effective and animal sparing colon specific bio-relevant dissolution media has been a foremost challenge facing pharmaceutical scientists over many decades.It is problematic to mimic the dynamic and ecologically diverse features of the colon in dissolution vessel.With the knowledge of enormous colonic microflora, the predominant species Bacteroides, Bifidobacterium, Eubacterium, Streptococcus and Lactobacillus species were cultured in 12% w/v skimmed milk powder and 5%w/v grade “A” honey. Probiotic culture was added to the dissolution media in order to test the drug release of polysaccharide based formulations. USP dissolution apparatus I/II with gradient pH dissolution method were used to evaluate the drug release from formulations meant for colonic drug delivery. Drug release from 5-fluorouracil granules and metronidazole tablets were assed under gastric, small intestine conditions and also within a simulated colonic environment involving existing rat caecal, human fecal media and compared with novel probiotic media. The present method can be successfully applied for the drug release testing of any oral formulations meant for colonic delivery.

Oral route is preferred for the treatment of various diseases/disorders. However, the oral bioavailability of nearly 40% of newly discovered molecules is diminished due to various physicochemical and physiological barriers. The reasons of low and variable oral bioavailability include, high lipophilicity, restricted permeability, first pass metabolism in the gut and liver and efflux transporter systems. These problems, in turn, lead to high intra- and inter-subject variability and lack of dose proportionality. To overcome these drawbacks, various formulation strategies have been adopted. However, the focus has recently shifted to lipid-based formulations owing to their ability to improve the oral bioavailability of poorly water soluble drug compounds. In this regard, self-emulsifying drug delivery systems (SEDDS), particularly nano-based systems, have been able to surmount the above mentioned problems. SEDDS are the recent technological innovations which are physically more stable and easier to manufacture with the globule size ranging between 10 to 1000 nm. Not only this, SEDDS are able to modulate the drug absorption leading ultimately to enhance oral bioavailability.

Conventional oral dosage forms provide a specific drug concentration in the systemic circulation without offering any control over the rate of drug delivery. Controlled-release drug delivery systems (CRDDS) provide drug release at a predetermined, predictable, and controlled rate. It was attempt to develop a gastro retentive drug delivery system of anti hypertensive drug using different polymers like that HPMCK4M, Xanthangum, PolyoxWSR303as matrixing agent, swelling agent, respectivel. A simplex lattice design was applied to combined effect of three formulation variables (ie, amount of HPMCK4M (X1), xanthangum (X2), and PolyoxWSR303 (X3). Results of multiple regression analysis indicated that low level of X1 and X 2 and high level of X3 should be used to manufacture the tablet formulation with desired in % swelling index and % drug release profile . When Polyox 303 was used, % swelling index increased, with increase on the total amount of drug released. Decreasing the amount of HPMC K4M while maintaining the total amount of polymer constant increased the release rate and total amount released. Decreasing the total amount of polymer decreases the swelling and work of penetration,

Most of the oral dosage forms possess several physiological limitations such as variable gastrointestinal transit because of variable gastric emptying leading to non-uniform absorption profiles, incomplete drug release and shorter residence time of the dosage form in the stomach. This leads to incomplete absorption of drugs having absorption window especially in the upper part of the small intestine, as once the drug passes down the absorption site, the remaining quantity goes unabsorbed.Since many drugs are well absorbed in the upper part of the gastrointestinal tract, such high variability may lead to non-uniform absorption and makes the bioavailability unpredictable. Hence, a beneficial delivery system to control and prolong the gastric emptying time and can deliver drugs in higher concentrations to the absorption site to show local action in the stomach requires a specialized delivery system. So the purpose of this study was to lengthen the stay of drug in its favorable absorption area and prolonging the release of the drug by formulating it into an effervescent gastro-retentive drug delivery system.
