During my M.S. thesis work I was involved in a multistep synthesis of solid support bearing a modified nucleoside for automated synthesis of siRNAs. First, I developed a three-step synthesis of protected 5'-amino uridine from uridine. Next I prepared the 3'-succinate of 5'-amino uridine in a one-pot two-step synthesis and attached it to a solid support via a succinamide linker. I had analyzed and characterized all new compounds using NMR techniques. Others in our laboratory used this solid support for synthesis of amide-modified siRNAs. We anticipate that the amide linkages will facilitate crossing the cellular membrane and improve nuclease resistance, biodistribution and pharmacokinetics of modified siRNAs.

There is a growing interest in the use of nanoparticles modified with DNAs, viruses, peptides and proteins for the rational design of nanostructured functional materials and their use in biosensor applications. These noble metal nanoparticles/biomolecules conjugates have specific properties and therefore they are attractive materials for nanotechnology in biochemistry and medicine. In this book, the author review work performed dealing with the DNA structure and functionalities, interactions between DNA, noble metal nanoparticles, surface active agents, solvents and other additives. Particular attention is given to how the DNA’s chain length and the DNA conformation affect the interaction and structure of the nanoconjugates and nanostructures that are formed. Also discussed are the recent advances in the preparation, characterization, and applications of noble metal nanoparticles that are conjugated with DNA aptamers and oligomers. Various government agencies around the world have proposed production of biodiesel as a renewable alternative to fossil fuel. Jatropha curcas is frequently mentioned as the best option for producing biodiesel. Despite of having potential as an alternative fossil fuel, J. curcas is not being fully exploited. Hence, there is a need to identify high yielding clones of J. curcas for its further improvement. Molecular marker analysis in genome studies enhance the speed and efficiency of crop improvement, for that we need the protocols of DNA isolation; which is used to obtain high quality and quantity of DNA. Here the objective is to carry out comparative analysis of nine different DNA isolation protocols, from those only six methods were able to isolate DNA from such secondary metabolite producing plant, J. curcas. The most effective method of DNA isolation is modified CTAB method. The extracted DNA from the leaves of Jatropha curcas had 83% purity. The extracted DNA was found suitable for restriction digestion and for different DNA fingerprinting techniques like, RAPD; which helps for the further improvement of plant. Doctoral Thesis / Dissertation from the year 2010 in the subject Biology - Micro- and Molecular Biology, grade: A, , course: Molecular Plant-microbe interaction - Agriculture/Rice Blast Disease/pathogenic shift/DNA double strand break repair, language: English, abstract: The ortholog of the Saccharomyces cerevisiae recombinational repair gene RAD51, Rhm51 was cloned from the genome of rice blast fungus Magnaporthe oryzae in order to clarify its role in the variability and pathogenicity. Rhm51 is a single copy gene, and is constitutively expressed at low levels during the cell cycle and at higher levels when the pathogen is grown in media supplemented with DNA damaging agents. We disrupted Rhm51 in two different strains of M. oryzae and both mutants showed reduced growth with conidia and colonies being highly sensitive to DNA double strand break-inducing agents. In addition, they lost the ability to undergo homologous recombination in adeA gene targeting. Mutants showed reduction in conidiation compared to the wild-type strains. Although mutants infected compatible rice cultivars, they produced significantly smaller and fewer lesions compared to wild-type strains and this might be due to the reduction in the amount of appressoria formed. DNA double-strand breaks (DSBs) quantified by the comet assay showed that loss of Rhm51 resulted in the accumulation of DSBs in mutant. With the ubiquity of powerful, mobile computers and rapid advances in sensing and robot technologies, there exists a great potential for creating advanced, perceptual computing environments. In this work, we investigate techniques for integrating passive, vision-based sensing into such environments, which include both conventional interfaces and large-scale environments. We discuss a methodology for vision-based human-computer interaction called the Visual Interaction Cues (VICs) paradigm. In the VICs paradigm, users interact with the computer system through natural hand gestures. We capture a "language of interaction" in which individual gestures are the words and sequences of gestures are the sentences. We give examples of both individual gestures and the gesture language, and present them on our next-generation interaction platform, the 4D Touchpad. Ultimately, our work takes a concrete step toward perceptual user interfaces.
